Learn how centrifugal partition chromatography (CPC) achieves unparalleled purity and yield in a single step, promising industrial-scale production of oligonucleotides for oligonucleotide development crucial in vaccine development and pharmacology.

This study was conducted to purify nucleotides, specifically a 20-mer single-stranded unmodified oligonucleotide, initially possessing 88.5% purity. Employing CPC, a two-phase solvent system of ethyl acetate/n-butanol/nuclease-free water was utilized. This approach yielded a remarkable purity of 96.6% and a calculated yield of 94.5%. Furthermore, the method enabled removal of organic solvents through lyophilization, with residual salts and additives easily eliminated via alcohol precipitation, which resulted in a pure and ready-for-application material. About CPC

CPC is a liquid-liquid preparative chromatographic technique, in which separation occurs between two immiscible liquid solvent systems: acting as both the stationary and mobile phases. The liquid stationary phase is immobilized by a strong centrifugal force inside cells attached to a rotor, and the mobile phase is then pumped through the stationary phase in the form of tiny droplets. Separation of the injected sample is based on the sample component’s varying partition coefficients between the mobile and stationary phases.
