The GMP-rCPC is a pilot-scale centrifugal partition chromatography (CPC) platform built for high-purity liquid–liquid purification. It bridges the gap between method development and commercial production, delivering validated performance, and full GMP compliance in a single system. Whether your workflow requires small-batch, campaign-style research or continuous manufacturing production, the system offers the flexibility and throughput to meet your operational needs. The system with the solvent handling modules as turn-key add-ons, is fully customizable for isolation, separation, purification, and remediation tasks further simplifying daily operation. The platform also enables seamless scalability from method development to production-level purification without re-engineering across your downstream workflow.
High Throughput, Outstanding Purity and Yield
With a 2-liter rotor, and throughput up to 2 kg/day, the GMP-rCPC delivers exceptional productivity in a compact footprint. The system is fully customizable and RotaChrom also offers custom solvent handling modules — including semi-automated preparation and recovery units — as turn-key add-ons for isolation, separation, purification, and remediation tasks, further simplifying daily operation.
Flexible Scale-Up and Scale-Out Without Re-Engineering
Boost productivity by running multiple rCPC units in parallel significantly increasing batch productivity, or achieve uninterrupted purification by operating two units in a loop for continuous processing for minimizing downtime, footprint and increasing operational efficiency. RotaChrom’s Adaptive Control System ensures synchronized, efficient performance in any configuration. As all RotaChrom systems share the same core CPC technology, GMP-rCPC ensures seamless technology transfer without the need for re-validation or re-engineering, if you scale from R&D or to full-scale production.
Built for Regulatory Compliance and Industry Standards
The system complies with cGMP, Eudralex Vol. 4, 21 CFR Part 11, GAMP 5, and supports EN 10204 3.1 material traceability. All wetted parts are FDA-grade, and the system holds CE, UKCA, and UL/CSA/NRTL certifications.
Chromatography From Research to Industry
Effortless scale-up from mg to kg/day to promote continuous production of APIs and macromolecules by liquid-liquid based centrifugal partitional chromatography (CPC). Select between lab-scale, pilot-scale, industrial-scale purification, or continuous manufacturing.
Versatile Purification Systems for a Wide Range of COIs
Purify macromolecules, peptides, nucleotides, steroids, stereoisomers, high or low polar compounds, drug-like compounds, and natural compounds.
Whether it's separation, isolation or remediation, the CPC’s extremely flexible and powerful solvent recycling system can be optimized to handle all of them according to the client request.
Purification Without silica gel
No more expensive silica gel or resign are needed. No irreversibly bounds, no silica degradation. Reach lower OPEX, lower total cost of ownership: Our Zero Silica Gel principle means that by applying CPC technology the total consumables can be substantially reduced and could be even decimated if a RotaChrom’ tailor-made solvent recovery system is in place.
Bespoke Design for Pharma
No two Partners are the same. Chromatographic challenges and goals, and facilities are all individual and unique, and cannot be handled by generic “cookie cutter” solutions. RotaChrom’s team can help its partners benefit from bespoke and custom-made purification solutions. Our team will ensure that the device the Partner receives is optimized so they can achieve maximum extraction.
In the field of downstream processing and bio-manufacturing, Centrifugal Partition Chromatography (CPC) is a versatile and highly efficient separation method. It provides high-resolution separation, exceptional productivity, and superior purity for binary mixtures while also offering a unique solution for the separation of chiral compounds, lipid nanoparticles, oligonucleotides, and proteins.
Oligonucleotide isolation
For industrial-scale production of oligonucleotides crucial in vaccine development and pharmacology, CPC can do it in a single purification step.
Purification of isomers
CPC can purify chiral compounds, epimers and diastereomeres (e.g. remdesivir). Purification can easily be scaled up to industrial-scale compared tot raditional chromatographic solutions.
Lipid nanoparticle purification
Lipid nanoparticles (LNPs) are crucial components in mRNA vaccine formulation in pharmaceutical drug delivery research, consisting of key lipid components. CPC can not only ensures the stability of the target lipid but also outperforms traditional chromatography techniques in terms of purity and yield.
Alkaloid remediation
Flow chemistry
Flow chemistry, also referred to as continuous flow, involves conducting chemical reactions within a continuous flow stream. CPC can be integrated into a downstream purification process needed for a flow chemistry operation, and the system can benefit from the advantages of both constituents.
CPC is a liquid-liquid preparative chromatographic technique, in which separation occurs between two immiscible liquid solvent systems: acting as both the stationary and mobile phases. The liquid stationary phase is immobilized by a strong centrifugal force inside cells attached to a rotor, and the mobile phase is then pumped through the stationary phase in the form of tiny droplets. Separation of the injected sample is based on the sample component’s varying partition coefficients between the mobile and stationary phases. CPC has several advantages like low operational expenses, easy scalability, elimination of sample loss and irreversible adsorption, operation without silica gel or any solid stationary phase, and the possibility to separate a wide array of compounds.
